ДЕЙСТВИЕ ЭКЗОПОЛИСАХАРИДОВ МОЛОЧНОКИСЛЫХ БАКТЕРИЙ НА ПРОЦЕСС ЗАЖИВЛЕНИЯ ОЖОГОВ У КРЫС

Цель исследования – изучить влияние экзополисахаридов (ЭПС) молочнокислых бактерий Lactococcus lactis В-1662 и Streptococcus thermophilus на заживление ожогов у самок белых крыс.

Материалы и методы. Использовали ЭПС Lactococcus lactis В-1662 и Streptococcus thermophilus, 5 % декспантенол. Животные (самки белых крыс), взятые для эксперимента, были разбиты на 5 групп: 1-я группа – без ожога, 2-я группа – с ожогом, 3-я группа – ожог и коммерческий препарат 5 % декспантенол, 4-я группа – ожог и 0,6 % раствор ЭПС L. lactis B-1662, 5-я группа – ожог и 0,6 % раствор ЭПС S. thermophilus. Ожог моделировали под эфирным наркозом на межлопаточном пространстве крысы дном пробирки, наполненной кипящей водой (2/3 объема) в течение 30 с (ожог степени IIIа). Для определения микрофлоры брали смывы с поверхности ожогов в течение всего эксперимента (28 сут).

Результаты. У животных с ожогом (2-я группа) через 10 сут наблюдали сокращение площади корки, а полное заживление ожога и восстановление шерстного покрова происходило через 28 сут. В 3-й группе крыс площадь ожоговой корки начинала уменьшаться только с 14-х сут, однако полное восстановление шерстного покрова наблюдали через 25 сут. У крыс 4-й и 5-й групп, леченых ЭПС L. lactis B-1662 и S. thermophilus, значительное сокращение площади ожога наблюдали уже на 3-и и 1-е сут соответственно с окончательным заживлением к 23-м сут. Изучение микрофлоры показало, что количество мезофильных аэробных и факультативных анаэробных микроорганизмов в 5-й группе крыс, раны которых обрабатывали ЭПС S. thermophilus, к моменту заживления раны было в 1,25 раза меньше по сравнению с другими группами крыс. При обработке ран ЭПС L. lactis B-1662 и S. thermophilus отмечали также значительное уменьшение количества бактерий группы кишечной палочки и стафилококка относительно других групп.

Выводы. Таким образом, ЭПС Lactococcus lactis В-1662 и Streptococcus thermophilus способствуют заживлению ожогов степени IIIа у крыс с полным восстановлением кожно-шерстного покрова, при этом отмечается значительное уменьшение числа бактерий группы кишечной палочки и стафилококков. Наилучший регенерирующий эффект выявлен в отношении ЭПС S. thermophilus.

EFFECT OF EXOPOLYSACCHARIDES OF LACTIC ACID BACTERIA ON BURN HEALING IN RATS

The aim of the paper is to study the effects of exopolysaccharides (EPS) of lactic acid bacteria Lactococcus lactis B-1662 and Streptococcus thermophilus on burn healing in female white rats.

Materials and Methods. We used EPS of Lactococcus lactis B-1662 and Streptococcus thermophilus, 5 % dexpanthenol. Experimental animals (female white rats) were divided into 5 groups: Group 1 – without burns, Group 2 – with burns, Group 3 – burns treated with 5 % dexpanthenol, Group 4 – burns treated with 0.6 % solution of EPS of L. lactis B-1662, Group 5 – burns treated with 0.6 % solution of EPS of S. thermophilus. The 3rd-degree burns were simulated under ether anesthesia on the interscapular space with the test glass filled with boiling water (2/3 volume; 30 seconds). To determine the microflora, washings were taken daily from the burn surface during the experiment (28 days).

Results. The animals belonging to Group 2 demonstrated the reduction of the crustlike surface of a healing skin lesion in 10 days, but the complete burn healing and restoration of the body hair coat was observed only after 28 days. In rats belonging to Group 3, the reduction of the crustlike surface was marked only in 14 days, however, the complete restoration of the body hair coat was observed after 25 days. In rats belonging to Groups 4 and 5, treated with EPS of L. lactis B-1662 and S. thermophilus, a significant reduction in the burn area was observed as soon as the 3rd and the 1st days. Complete healing was noticed by the 23rd day. The study of microflora showed that the number of mesophilic aerobic and facultative anaerobic microorganisms in Group 5 (the wounds were treated with EPS of S. thermophiles) by the time of wound healing was 1.25 times as small as in other groups of rats. Treating wounds with EPS of L. lactis B-1662 and S. thermophilus also led to a significant decrease in the number of Escherichia coli bacteria and staphylococcus in comparison with other groups.

Conclusion. Thus, exopolysaccharides of Lactococcus lactis B-1662 and Streptococcus thermophilus contribute to the 3rd-degree burn healing in rats, to complete restoration of cutaneous and body hair coat, and to significant decrease in the number of Escherichia coli and Staphylococcus bacteria. The best regenerating effect was revealed while treating rats with EPS of S. thermophilus.
